双水相亲和萃取法从豆壳中分离过氧化物酶

摘要: 采用双水相金属螯合亲和萃取法从豆壳中分离过氧化物酶,考察了豆壳过氧化物酶在不同类型双水相系统中金属螯合亲和配基聚乙二醇(PEG)-亚氨基二乙酸(IDA)-Cu(Ⅱ)对亲和分配的影响,发现PEG/羟丙基淀粉(PES)系统适合于金属螯合亲和分配豆壳过氧化物酶。进一步考察亲和配基加入量、成相聚合物分子量、系统组成、pH等因素对PEG/PES系统亲和分配的影响,优化分离条件为:PEG20009%,PES20014%,PEG-IDA-Cu(Ⅱ)1%,豆壳过氧化物酶的分配系数达40,纯化因子为2.8,回收率达到93%。

Abstract: Metal chelating affinity partition with aqueous two-phase system was studied to separate the peroxidase fromsoybean hull extract. The effects of affinity ligand (polyethylene glycol (PEG)- iminodiacetic acid (IDA)-Cu(II)) on the affinitypartition of soybean hull peroxidase (SHP) were investigated in different types of aqueous two-phase systems. The resultsindicated that PEG/ hydroxypropyl starch (PES) aqueous two-phase system was suitable for affinity extraction of SHP. Severalfactors, such as adding amount of affinity ligand, molecular weight of phase-forming polymer, system compositions, pH, etc.,were optimized. The best separation conditions were: PEG2000 9%, PES200 14% and PED-IDA-Cu(II) 1%. The partitioncoefficient of SHP could reach 40, and purification factor was 2.8 with the recovery of 93%.
