大孔吸附树脂分离纯化杜仲绿原酸研究

Study on Separation and Purification of Chlorogenic Acid from Eucommia ulmoidues by Macroporous Resin

摘要: 以杜仲叶60%乙醇提取液为原料,研究了NKA-2、NKA-9、LS600、HZ806、HZ906、HPD100、HPD500、S-8八种大孔吸附树脂对绿原酸的静态吸附与解吸效果,优化了最优树脂分离纯化杜仲叶绿原酸的工艺技术参数。结果表明,NKA-2树脂对绿原酸具有较好的吸附与解吸性能,在pH为3的条件下静态吸附量为86mg/g,解吸率达93.23%;当主要考虑绿原酸得率时,最优工艺参数为上样浓度4mg/ml、上样流速4BV/h、上样体积5BV、洗脱剂乙醇浓度80%、洗脱流速为3BV/h、洗脱体积为2BV,在该工艺参数下,绿原酸得率为83.14%,纯度为28.91%;当主要考虑绿原酸纯度时,最优工艺参数为上样浓度4mg/ml、上样流速4BV/h、上样体积5BV、洗脱剂乙醇浓度80%、洗脱流速为3Bv/h、洗脱体积为1.5Bv,在该工艺参数下,绿原酸得率为79.62%,纯度为39.38%。

Abstract: Studies on separating of chlorogenic acid by macroporous resins,the material is 60% ethanol extracts of eucommia leaves. The research compared the absorption capacity and desorbtion effect with different concentration of alcohol of 8 kinds of resins :S-8、HZ-806、HZ-906、HPD-500、 HPD-100、NKA-2、NKA-9 and LS-600,and the best processing parameters were optimized based on the adsorption and desorption results. The results showed that NKA-2 was the best one. Its rate of the static absorbtion was 83mg/ml and the rate of desorption was 93.23% when the pH is 3. An optimum yield parameter with sample concentration 4mg/ml, sample flow rate 4BV/h , sample volume 5BV, elution ethanol concentration 80%, elution flow rate 3.0BV/h, elution volume 2BV, was built and applied to separate chlorogenic acid ,in this situation the result was obtained that yield is 83.14% and the purity is 28.91%..An optimum purification parameter with sample concentration 4mg/ml, sample flow rate 4BV/h , sample volume 5BV,elution ethanol concentration 80%, elution flow rate 3.0BV/h, elution volume 1.5BV, was built and applied to separate chlorogenic acid ,in this situation the result was obtained that yield is 79.62% and the purity is 39.38%.
