苹果皮黄酮对D-半乳糖致衰老模型小鼠抗氧化能力的影响

摘要: 摘 要:目的 探究苹果皮总黄酮(Flavonoids from Apple Pericarp,FAP)对D-半乳糖致衰小鼠的抗衰老作用。方法 采用皮下注射D-半乳糖制备衰老小鼠模型。筛选 50 只健康昆明小鼠(雌雄各半)随机分为模型组、空白对照组和低、中、高剂量实验组。于造模第16天后,给低、中、高剂量组小鼠分别灌胃50mg/kg、100mg/kg、200mg/kg苹果皮总黄酮,持续45天后,采用Morris水迷宫和小鼠避暗仪测试小鼠学习记忆能力,并采集血液、取脑、肝、心、肾组织,测定各组织MDA含量、SOD活性及GSH活力。结果 与正常对照组相比,模型组小鼠学习记忆能力减退,除心之外,脑、肝、肾、血液MDA含量显著增加(P<0.05),SOD活性和GSH活力显著下降(P<0.05);与模型组相比,除低、高剂量组的血液SOD活性与GSH-Px活力及低剂量组心脏GSH-Px活力无显著差异性外(P>0.05),其他各组织心、脑、肝、肾SOD活性与GSH-Px活力均有显著差异性(P<0.05),但各剂量组与正常对照组相比,中剂量组各指标均无显著差异性(P>0.05),而低、高剂量组MDA含量显著高于正常对照组(P<0.05),SOD活性和GSH活力显著低于正常对照组(P<0.05)。 结论 经口给予小鼠一定剂量的苹果皮黄酮有明显的抗衰老的作用,且有剂量效应。

Abstract: Abstract:Objective To study the antioxidant effects of the total flavonoids from apple pericarp in D-galactose-induced aging mice. Method Hypodermic injection of D-galactose were used to set up the aging model. Fifty physical health Kunming mice were choosed and randomly divided into five groups (n=10,half male):normal control group, model group, low (50mg/kg),middle (100mg/kg) and high dose (200mg/kg) of experimental group (EG) of total flavonoids administered groups. Model and EG group mice were hypodermicly injected by D-galactose-induced for 45 days.The EG were intragastric administrated the total flavonoids from the sixteenth days after modeling.The learning and memory ability in mice were test using the dark-avoided meter and Morris water maze method on the forty-sixth day.Then, the mice were killed, blood, brain, liver, heart, kidney tissue were collected, and the indexes were determined of tissues MDA contents, SOD activities and GSH activities. Result Compared with the normal control group, the learning and memory ability were decreased of model group mice. the MDA content of brain, liver, kidney and blood were increased significantly(P<0.05)except heart tissue ,SOD and GSH activity were decreased significantly(P<0.05);Compared with the model group, the SOD and GSH ability were significant deviation(P<0.05)but the SOD and GSH of blood in low and high does of group and the GSH of heart in low group(P>0.05). Compared with the normal control group,there were not significant deviation in middle dose of group mice (P>0.05).The MDA in low and high does of group mice were improved significantly(P<0.05) while the SOD and GSH activity were decreased significantly(P<0.05)of high does of group’s. Conclusion The mice intragastric administrated the total flavonoids have antioxidant effect on the memory of aging mice, but the dose should be controled in a certain extent.
