纳豆冻干粉对抗生素介导小鼠免疫调节作用和细胞因子分泌的影响

Effects of Natto lyophilized powder to antibiotics mediated immunomodulatory and the secretion of cytokines in mice

摘要: 本试验旨在探究纳豆冻干粉(Natto lyophilized powder,NLP)对SPF小鼠免疫功能及其细胞因子分泌的影响。选取体质健康小鼠128只,随机分为8组:空白对照组C,调节组R共4组,预防组P共2组,模型组M。除C组外,灌胃3 d抗生素溶液建模,R组分别灌胃生理盐水和三个剂量组的NLP溶液,P组每天继续灌胃抗生素溶液8?h后分别灌胃生理盐水和NLP溶液,M组灌胃抗生素溶液,灌胃30 d后测定免疫指标及其细胞因子分泌的含量变化情况。结果表明:与模型组相比,低剂量的NLP能够极显著增加脾脏和胸腺指数(p<0.01),NLP调节组能够极显著增加血清中IL-2,IL-10,TNF-α,IFN-γ的分泌量(p<0.01), NLP能够极显著增加iNOS活力和小鼠血清中的白蛋白、球蛋白和白球比的值(p<0.01),低剂量和高剂量NLP调节组能极显著增加NO水平(p<0.01)。试验组的小鼠血清溶血素水平较对照组极显著升高(p<0.01)。NLP均能够显著增强小鼠腹腔巨噬细胞吞噬作用(p<0.05)。与空白对照组相比,NLP调节组的半数溶血值显著升高 (p<0.05)。NLP高剂量调节组和中剂量预防组极显著增强巨噬细胞的吞噬功能(p<0.01),极显著增加巨噬细胞吞噬百分率、鸡红细胞的吞噬数(p<0.01)。高剂量NLP调节组iNOS的活力和白蛋白的含量极显著升高(p<0.01),中剂量的NLP预防组的白蛋白和球蛋白的含量升高。结果:NLP具有免疫增强作用和增加细胞因子分泌的作用。

Abstract: The natto lyophilized powder (NLP) to antibiotics mediated immunomodulatory effects and the influence of the secretion of cytokines in SPF mice were studied in this paper.128 physique healthy mice were selected and divided randomly into 8 groups: control group C, four regulation groups R, two prevention groups P, model group M. In addition to the group C, antibiotics solution was lavaged for 3 d for modeling. Saline and three dose of NLP solution were lavaged respectively in R group. Saline and NLP solution were lavaged respectively after antibiotics solution lavaged for 8 h in P group. Antibiotics solution was lavaged in M group. Immunomodulatory effects and the influence of the secretion of cytokines in the content variation of immune index and the secretion of cytokines were determined after lavaged for 30 days. Results show that , compared with model group,low dose of NLP of spleen and thymus index was extremely significantly higher than model group. The production of serum IL-2, IL-10, TNF-alpha, IFN-gamma were significantly increase in NLP regulation group compared with model group (p< 0.01) .The iNOS activity and the albumin, globulin and globulin ratio in mice serum and the phagocytizing effect of the chicken red blood cell by peritoneal macrophage(PM) in experimental group was significantly higher than model group. The NO level in high and low doses of NLP regulation group was significantly higher than model group. The mice serum hemolysin level in experimental group was significantly higher than control group(p<0.01). The spleen lymphocyte transformation factor levels in experimental groups were significantly increased compared with the model group(p<0.05). Compared with the blank control group, the half hemolysis value of NLP adjust group (p<0.05) was significantly increased. The phagocytosis function of macrophage was significantly enhanced (p<0.01) and the phagocytosis percent of macrophages and red blood cell count of chicken was significantly increased (p<0.01). The iNOS activity and the content of albumin were significantly increased in high dose of NLP adjustment group(p<0.01), the albumin and globulin content in middle dose of NLP prevention group was increased. Results: NLP can enhance immune function and increase the secretion of cytokines.
