姜黄素对幽门螺杆菌及其诱导的人胃GES-1细胞损伤的影响

Effect of Curcumin on Helicobacter pylori and Helicobacter pylori Injured GES-1 Cells

摘要: 本文研究姜黄素对幽门螺杆菌(Helicobacter pylori, Hp)和脲酶的抑制作用,及对正常GES-1细胞和Hp感染GES-1细胞的影响。抑菌实验结果显示,姜黄素可明显抑制Hp生长,其最低抑制浓度为200 μM。姜黄素对脲酶活性也有明显抑制作用,其半数抑制浓度IC50为1.735 mM。短时间内(12 h)较低浓度(68 μM)姜黄素对正常GES-1细胞增殖无明显影响,但姜黄素浓度和作用时间的增加可使细胞存活率明显下降。经68 μM姜黄素作用12 h后,损伤模型细胞形态有一定程度的复原,细胞存活率略有升高,但无统计学意义;高浓度(136、680 μM)姜黄素则会导致细胞存活率明显下降。因此,姜黄素对Hp和脲酶有明显的抑制作用,但不足以缓解Hp对GES-1细胞的损伤作用,还需配合具有促进胃黏膜细胞增殖的活性物质协同作用。

Abstract: The inhibition of curcumin on Helicobacter pylori (Hp) and jack bean urease activity was studied. The effect of curcumin on normal GES-1 cells and Hp induced GES-1 injury was also researched. The results of bacteriostatic experiments showed that curcumin significantly inhibited Hp proliferation, with the MIC of 200 μM. Moreover, curcumin showed obvious inhibitory effect on jack bean urease activity, with the IC50 value of 1.735 mM. Curcumin could inhibit human gastric epithelial GES-1 cells in a dose- and time-dependent manner. After infecting GES-1 cells with Hp for 24 h, the GES-1 cells injury model was successfully established to study the effect of curcumin on injured GES-1 cells. After treatment with curcumin at the concentration of 68μM for 12 h, the injury cell model morphology was slightly restored , the survival rate of GES-1 cells slightly increased. But the survival rate significantly decreased after treatment with curcumin at the concentration of 136 μM or 680 μM. Thus, inhibition of curcumin on Hp and jack bean urease could not obviously help to repair Hp induced GES-1 cells damage. It may be helpful to reduce Hp induced GES-1 cells damage with the synergistic effects between curcumin and other bioactive ingredients with promoting effect on GES-1 cells proliferation.
