摘要: 将鼠抗Cd2+-IEDTA单克隆抗体和鼠抗Pb2+-IEDTA单克隆抗体分别与包载CdSe/CdS量子点(quantum dots,QDs)的聚苯乙烯微球(polystyrene,PS)偶联,建立一种可快速检测铅镉的量子点荧光定量串联卡及免疫层析竞争检测方法。串联卡的最佳反应时间为5 min,可检测的Pb2+和Cd2+浓度范围分别为1-500 ng/mL和1-400 ng/mL,检测变异系数(coefficient of variance,CV)小于10%;检测特异性良好,与其他金属离子无明显交叉反应;利用该方法与电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry,ICP-MS)进行了肉类食品检测对比,检测结果与ICP-MS相关性良好。该检测方法为同时定量检测铅镉提供了快速简便的有效途径,适用于监测肉中Pb2+和Cd2+含量是否超标。

Abstract: The object of this study was to establish a quantitative dual fluorescence immunochromatographic assay for rapid detection of lead and cadmium, which is an immunochromatographic competitive detection method based on quantum dots. Mouse anti-Cd2+-IEDTA and anti-Pb2+-IEDTA monoclonal antibody were coupled with CdSe/CdS core-shell quantum dots embedded in polystyrene. The optimum reaction time of the dual detection strip was 5 min. The linear range of Pb2+ detection was 1-500 ng/mL, and the linear range of Cd2+ detection was 1-400 ng/mL. The coefficient of variations of this dual assay were less than 10% for both lead and cadmium. Moreover, there was no cross-reactivity with other metal ions. The results of this assay were compared with inductively coupled plasma mass spectrometry measuring meat samples purchased from markets, and the dual assay correlated well with standard method. This dual fluorescence immunochromatographic assay provided an applicable method for rapid and quantitative detection of lead and cadmium simultaneously, which is suitable for monitoring whether the content of Pb2+ and Cd2+ in meat exceeds the standard.
