摘要: 目的:建立铁皮石斛饮料中D-甘露糖(D-Man)含量的测定方法。方法:样品分别经乙醇沉淀、盐酸水解和衍生化处理。色谱柱使用Agilent EclipseXDB-C18(4.6mm×250mm,5μm); 20mmol/L乙酸铵-乙腈溶液(80.5:19.5)为流动相;流速1.0mL/min;检测波长250nm。结果:样品中西洋参多糖对甘露糖含量测定无干扰;样品中D-甘露糖在7.815~312.6μg/mL 范围内,线性关系均良好,相关系数r=0.9997;样品回收率为99.66%,RSD为1.76%(n=6)。结论:本方法专属性强、灵敏、精确、重现性好,可用于本品多糖组成中D-Man含量的测定。

Abstract: Objective: To establish a HPLC method to determine the content of D-mannose (D-Man) in Dendrobium officinale Kimura et Migo beverage (DOB). Methods: Ethanol precipitation followed by hydrochloric acid hydrolysis and derivatization was carried out for sample preparation. Mannose was separated on an Agilent Eclipse XDB-C18 (4.6 mm × 250 mm, 5μm) using a mobile phase made up of 20 mmol/L ammonium acetate and acetonitrile (80.5:19.5, V/V) at a flow rate of 1.0 mL/min. The detection wavelength was set at 250 nm. Results: No interference was found from Panacis quinquefolium in DOB. The linear range of Man was between 7.815μg/mL and 312.6μg/mL (r = 0.9997). The average recovery was 99.66% (RSD 1.76%, n = 6). Conclusion: This method is specific, sensitive, accurate and reproducible and may therefore be applicable for the determination of D-Man in DOB.
