裙带菜多糖体外和体内降血脂活性

In Vitro and in Vivo Hypolipidemic Effect of Undaria pinnatifida Polysaccharide

摘要: 目的:采用体外和体内法研究裙带菜多糖(Undaria pinnatifida polysaccharide,PUP)降血脂功能。方法:模拟人体胃肠环境,体外法测定PUP对牛磺胆酸盐的结合能力及其对胰脂肪酶的抑制作用,以此来评价其体外降血脂功能;通过高脂饮食诱导C57BL/6J小鼠建立高脂血症模型,利用PUP灌胃干预,比较PUP低、高剂量组与高脂血症组的小鼠体质量、血清血脂四项含量、血清抗氧化指标、血清肝标记酶的活力、肝脏组织固定切片等指标。结果:体外模拟降血脂实验结果表明,10 mg/mL PUP对牛磺胆酸盐的结合率可达到52.3%,1.0 mg/mL PUP对胰脂肪酶活力的抑制率为31.9%,说明PUP有较好的体外降血脂作用;体内降血脂动物实验结果表明,PUP能抑制高脂饮食小鼠体质量的增长,抑制因高脂饮食导致的血清总胆固醇、甘油三酯、低密度脂蛋白胆固醇水平的增加,升高高密度脂蛋白胆固醇的水平,提高小鼠抗氧化体系相关物质的水平,降低丙二醛的水平,降低血清肝标记酶的活力,减轻高脂饮食导致的肝脏损伤。结论:体外和体内实验表明PUP具有较好的降血脂活性。

Abstract: Objective: In vitro and in vivo methods were used to explore the hypolipidemic effect of Undaria pinnatifida polysaccharide (PUP). Methods: In a simulating gastrointestinal environment, the taurocholate-binding capability of PUP and its inhibitory effect on pancreatic lipase were determined. C57BL/6J mice were fed a high-fat diet (HFD) to induce hyperlipidemia. The hyperlipidemic mice were gavaged with simvastatin, physiological saline or PUP at low and high doses. Body mass, four serum lipid parameters, serum antioxidant indices and serum and liver marker enzyme activities as well as hepatic pathological changes in the PUP groups and the hyperlipidemia group were measured and compared with each other. Results: At a concentration of 10 mg/mL, the binding rate of PUP to taurocholate was 52.3%, and the inhibition rate of pancrelipase by PUP at 1.0 mg/mL was 31.9%. In addition, PUP alleviated body mass gain in HFD-fed mice, suppressed the increase in total triglycerides (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), increased the level of high density lipoprotein cholesterol (HDL-C), and improved the antioxidant status. PUP also reduced the content of malondialdehyde (MDA), the activity of serum and liver marker enzymes and liver damage caused by HFD. Conclusion: PUP has a significant hypolipidemic effect in vitro and in vivo.
