刺梨果多糖对异丙肾上腺素诱导的心肌缺血小鼠保护作用及机制

Protective Effect and Mechanism of Rosa roxburghii Tratt Polysaccharides on Isoprenaline-Induced Myocardial Ischemia in Mice

摘要: 目的:探究刺梨果(Rosa roxburghii Tratt)多糖对异丙肾上腺素(isoprenaline,ISO)诱导的心肌缺血(myocardial ischemia,MI)小鼠保护作用及作用机制。方法:将40 只雄性C57BL/6小鼠随机分为5 组:对照组,模型组,普萘洛尔(30 mg/kg)组,刺梨果多糖低、高剂量(50、100 mg/kg)组,每组8 只。各给药组小鼠连续灌胃(ig)给药14 d,从第8天开始,ig给药1 h后,除对照组腹腔注射(ip)相同剂量的生理盐水外,其余各组小鼠ip ISO(10 mL/kg),连续7 d,构建MI小鼠模型。末次给药1 h后检测小鼠心电图变化,麻醉后取心脏,计算心脏指数;采用苏木素-伊红(hematoxylin-eosin,HE)染色观察心肌组织病理变化;采用2% 2,3,5-氯化三苯基四氮唑染色液(2,3,5-triphenyltetrazolium chloride,TTC)染色检测心肌梗死面积;采用生化试剂盒检测小鼠血清中乳酸脱氢酶(lactate dehydrogenase,LDH)活性、超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量;采用免疫印迹法(Western blot)检测心脏组织中NADPH氧化酶2(NADPH oxidase 2,NOX2)、B细胞淋巴瘤2(B-cell lymphoma2,Bcl2)、Bcl2相关X(Bcl2-associated X,Bax)、半胱天冬酶3(Caspase 3)、中核因子E2相关因子2(nuclear factor E2-related factor 2,Nrf2)、Kelch样ECH相关蛋白1(Kelch-like ECH-related protein 1,Keap1)、血红素氧合酶1(heme oxygenase 1,HO-1)蛋白的表达情况。结果:与对照组小鼠比较,模型组小鼠心脏肿大,心电图ST段发生明显升高,HE染色可观察到心脏组织出现明显的炎症浸润和细胞间隙,TTC染色结果发现明显的心肌组织梗死。进一步实验发现模型组小鼠血清MDA含量和LDH活性显著升高(P<0.001),自由基清除酶SOD活性降低(P<0.05),Western blot结果显示,Nrf2、HO-1和Bcl2显著下调(P<0.05,P<0.01),Keap1、NOX2、Bax和Caspase 3显著上调(P<0.05,P<0.01)。与模型组小鼠比较,刺梨果多糖组小鼠心肌损伤都有不同程度的恢复,心电图ST段降低,小鼠心脏组织病理损伤减少,血清MDA含量和LDH活性显著降低(P<0.001),SOD活性显著升高(P<0.01,P<0.001),心脏组织蛋白Nrf2、HO-1和Bcl2表达显著上调(P<0.05,P<0.01),Keap1、NOX2、Bax和Caspase 3表达显著下调(P<0.05,P<0.01,P<0.001)。结论:刺梨果多糖能通过调节Keap1-Nrf2信号通路,抑制氧化应激损伤,继而调控Caspase 3、Bax、Bcl2蛋白,减少心肌细胞凋亡发生,从而对ISO诱导的MI小鼠起到保护作用。

Abstract: Objective: To explore the protective effect and underlying mechanism of Rosa roxburghii Tratt polysaccharides on isoproterenol (ISO)-induced myocardial ischemia (MI) in mice. Methods: Forty male C57BL/6 mice were equally and randomly divided into five groups: control, model, propranolol (30 mg/kg), low- and high-dose R. roxburghii Tratt polysaccharides (50 and 100 mg/kg). Mice were pretreated with R. roxburghii Tratt polysaccharides for seven days followed by intraperitoneal injection of isoprenaline (ISO, 10 mL/kg) or physiological saline for seven days. Electrocardiograms were recorded 1 h after the last injection. The mice were anesthetized and hearts were excised to calculate cardiac index. Hematoxylin-eosin (HE) staining was used to evaluate myocardial pathological changes. The area of myocardial infarction was detected by 2% 2,3,5-triphenyltetrazolium chloride (TTC) staining. Serum lactate dehydrogenase (LDH) activity, superoxide dismutase (SOD) activity and malondialdehyde (MDA) levels were detected by biochemical kits. The protein expression of NADPH oxidase 2 (NOX2), B-cell lymphoma 2 (Bcl2), Bcl2-associated X (Bax), caspase 3, nuclear factor E2-related factor 2 (Nrf2), Kelch-like ECH-related protein 1 (Keap1) and heme oxygenase 1 (HO-1) in myocardial tissues were detected by Western blot. Results: Compared with the control group, heart coefficient was significantly increased in the model group and the ST segment of electrocardiogram was significantly elevated. Significantly pathological changes and increased myocardial infarction were found in the myocardial tissue of mice in the model group. Serum MDA and LDH levels were increased (P < 0.001) and SOD activity was inhibited in the model group (P < 0.05). The protein expressions of Nrf2, HO-1 and Bcl2 were significantly down-regulated (P < 0.05 or P < 0.01), while those of Keap1, NOX2, Bax and caspase 3 were significantly up-regulated in the model mice (P < 0.05 or P < 0.01). The ST segment of electrocardiogram was lowered, and myocardial pathological changes were restored in the polysaccharide treatment group compared with the model group. In addition, serum MDA and LDH levels were significantly decreased (P < 0.001) and SOD activity was significantly increased (P < 0.01 or P < 0.001). The protein expressions of Nrf2, HO-1 and Bcl2 were significantly up-regulated (P < 0.05 or P < 0.01), while those of Keap1, NOX2, Bax and caspase 3 were significantly down-regulated (P < 0.05, P < 0.01 or P < 0.001). Conclusion: R. roxburghii Tratt polysaccharides ameliorate ISO-induced myocardial ischemia though reducing oxidative stress, regulating caspase-3, Bax and Bcl2 protein expressions and suppressing myocardial cell apoptosis via the Keap1-Nrf2 signaling pathway.
