新疆酸李提取物对脂多糖诱导RAW264.7炎症模型细胞氧化应激损伤的缓解作用及机制

Alleviative Effect and Mechanism of Prunus cerasifera Ehrhart Extract on Oxidative Stress Injury in Lipopolysaccharide-Stimulated RAW264.7 Cells

摘要: 本研究探讨新疆酸李(Prunus cerasifera Ehrhart)提取物对脂多糖(lipopolysaccharide,LPS)诱导RAW264.7炎症模型细胞的抗炎作用及其潜在的分子机制。首先,对新疆酸李提取物进行乙醇梯度洗脱,并分别检测不同体积分数乙醇溶液洗脱部位的多酚和黄酮含量以及抗氧化活性;其次,针对体积分数70%乙醇溶液洗脱组分(PCEE-70),分析其对LPS诱导的RAW264.7炎症细胞中抗氧化及氧化应激有关生化指标的影响;最后,通过蛋白免疫印迹(Western blot,WB)实验检测核因子κB(nuclear factor kappa B,NF-κB)p65和磷脂酰肌醇3-激酶(phosphoinositide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,AKT)/c-Jun氨基末端激酶(c-Jun N-terminal kinase,JNK)信号通路有关磷酸化及非磷酸化蛋白的表达水平。结果表明,PCEE-70相较于其他洗脱组分具有较高的多酚和黄酮含量,并具有较强的多种抗氧化活性。PCEE-70能明显降低LPS诱导的RAW264.7炎症细胞中NO、活性氧和丙二醛含量,提高乳酸脱氢酶、超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶活力,降低白细胞介素(interleukin,IL)-6、IL-1β和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)促炎因子的水平。此外,WB结果表明,PCEE-70能明显降低LPS诱导的RAW264.7炎症细胞中TNF-α、p-NF-κB p65、p-NF-κB抑制蛋白、p-PI3K、p-AKT及p-JNK蛋白的表达。综上,PCEE-70能抑制LPS诱导RAW264.7炎症细胞炎性因子的分泌,从而延缓氧化应激反应对细胞的损伤,其作用机制与抑制NF-κB和PI3K/AKT信号通路的激活有关。

Abstract: This study aimed to investigate the anti-inflammatory effects of Prunus cerasifera Ehrhart extract (PCEE) on lipopolysaccharide (LPS)-stimulated RAW264.7 cells and to elucidate its underlying molecular mechanism. First, PCEE was fractionated by gradient elution with ethanol, and the separated fractions were evaluated for total polyphenol and flavonoid contents, as well as antioxidant activity. Next, the 70% ethanol-eluted fraction (PCEE-70) was selected to measure its effects on biochemical indexes related to antioxidant defense and oxidative stress in LPS-stimulated RAW264.7 cells. Finally, Western blot (WB) was employed to detect the expression levels of phosphorylated and non-phosphorylated proteins associated with the nuclear factor kappa B (NF-κB) p65 and phosphoinositide 3-kinase/protein kinase B/c-Jun N-terminal kinase (PI3K/AKT/JNK) signaling pathways. The experimental results demonstrated that PCEE-70 had higher contents of polyphenols and flavonoids along with stronger antioxidant activities compared to the other fractions. PCEE-70 significantly inhibited the levels of nitric oxide (NO), reactive oxygen species (ROS) and malondialdehyde (MDA), while enhancing the activities of lactate dehydrogenase (LDH), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px). It also reduced the levels of the pro-inflammatory cytokines interleukin (IL)-6, IL-1β, and tumor necrosis factor-α (TNF-α). Furthermore, WB results indicated that PCEE-70 significantly downregulated the expression of TNF-α, p-NF-κB p65, p-IκB, p-PI3K, p-AKT, and p-JNK proteins. In conclusion, PCEE-70 can inhibit the secretion of inflammatory factors in LPS-induced RAW264.7 cells, thereby mitigating oxidative stress-induced cellular damage. Its action mechanism is associated with the suppression of NF-κB and PI3K/AKT signaling pathway activation.
