摘要: 本文研究了两种不同的连接方法在野油菜黄单胞菌(Xanthomonascampestris)xanA基因与pUCm-T载体连接中的连接效果,并进行了转化实验进行鉴定。实验结果表明,xanA基因与pUCm-T载体的适宜连接条件为16℃连接过夜。连接体系中,xanA基因与pUCm-T载体的体积比为4:1。

Abstract: Two methods for gene xanA to ligate with pUCm-T vector were discussed in this thesis. And the transformations were also done for testing. It showed that the better ligation condition was ligated at 16℃ for overnight. In the ligation system,the volume ratio of gene xanA to pUCm-T vector was 4:1.
