蝉拟青霉菌丝体糖蛋白的分离提取及组分测定

Extraction and Separation of Glycoproteins from the Mycelia of Paecilomyces cicadae with Analysis Test

摘要: 蝉拟青霉菌粉经脱脂、水提、丙酮沉淀、蒸馏水透析后得到糖蛋白提取物GP,经苯酚-硫酸法测定总糖含量为34.79%;Lowry法测定蛋白质含量为63.45%;糖蛋白提取物GP上SephdexG-25柱层析,逐管用紫外吸收法(280nm)检测,发现有两个峰(PS1和PS2)。PS1总糖含量为21.11%,蛋白质含量为77.1%,PS2总糖含量为44.98%,蛋白质含量为53.02%。

Abstract: By means of degreasing, water-extracting, acetone-precipitating and dialyzing in distilled water, the total carbohydrate in the mycelial powder of Paecilomyces cicadae is tested as 34.79% by Phenol-Sulphuric Measure method and the protein content is 63.45% by Lowry. The glycoprotein possesses two absorbing peaks (PS1 and PS2) under ultraviolet spectrum as separated by Sephadex G-25. The protein content is 77.1% in PS1 and 53.02% in PS2, while the total carbohydrate is 21.11% in PS1 and 44.98% in PS2, respectively.
