双酶法制备林蛙油小分子肽的研究

Study on Preparation of Micromolecule Peptide from Oviductus Ranae of Rana temporaria chensinensis David with Double Enzyme Method

摘要: 采用木瓜蛋白酶和碱性蛋白酶双酶结合法对林蛙油进行酶解,利用凝胶层析法和HPLC法进行检测。结果表明:木瓜蛋白酶的最佳酶解条件为底物浓度1%、酶用量1200U/g、温度60℃、时间3.5h、pH6.0,碱性蛋白酶的最佳条件为加酶量500U/g、温度50℃、时间3.0h、pH8.0。酶解液利用SephadexG-15柱分离得到分子量小于1355D的肽段,经HPLC分析检测,色谱图上出现了三个高低不同的峰,其保留时间依次为21.837、23.300和24.499min。同时,木瓜蛋白酶和碱性蛋白酶结合可以有效地将林蛙油蛋白降解为肽,水解度达到17.8%,比单酶提高了5.6%;且产物具有较好的溶解性和流动性。

Abstract: Oviductus Ranae of Rana temporaria chensinensis David was hydrolyzed by the combination of caroid and alkaline proteinase,and the gel chromatography and HPLC were employed for determination.The result showed that the optimal hydrolysis conditions by caroid are:substrate concentration 1%,enzyme dosage 1200 U/g,temperature 60 ℃,time 3.5 h and pH 6.0.By alkaline proteinase they are enzyme dosage 500 U/g,temperature 50 ℃,time 3.0 h,and pH 8.0.Molecular weight less than 1355 D peptid was obtained by Sephadex G-15 separating column,and three peaks appearanced in the chromatogram by HPLC detection,whose retention time was 21.837,23.300 and 24.499 min in turn.Meanwhile,the Oviductus Ranae protein can be effectively degradated into peptide by the combination of the two enzymes,and the degree of hydrolysis reaches 17.8%,increasing by 5.6% compared with single enzyme.The production dissolubility and flowability are both better.
