树舌灵芝菌丝体发酵工艺条件试验

Study on Fermentation Technical Conditions of Ganoderma applanatum Mycelia

摘要: 树舌灵芝是灵芝属真菌中的一种,是我国中医药宝库中的珍贵药用菌。为寻求高产的生长条件和经济的培养基,进行了转速、温度、pH值、装瓶体积、接种量、碳源、氮源、C/N等单因素试验和多因素试验设计,利用菌丝体液体培养探索这些条件对菌丝体产量的影响。结果表明,菌丝生长的最适转速为150r/min,最适温度为25~30℃,最适pH值为5.5~6.5,最适装瓶体积为125ml,最适接种量为5ml/100ml;最适碳源为葡萄糖,最适氮源为米糠,最适C/N为25:1。液体培养的最佳营养条件为:3%的葡萄糖,0.072%的KNO3,0.02%的NaH2PO4,pH为8.5。通过生长曲线实验得出logistic方程:Y=0.616/(1+17.732e-8.874x),菌丝体干重达到500~700mg/100ml。

Abstract: Ganederma applantatum is a species of the Ganoderma genus; it is one of the precious medicinal mushroom in the treasury of Chinese traditional medicines. To study the high yield and economical medium productive conditions, this experiment selected and used single factors such as rational speed, temperature, pH value, bottled volume, inoculated bulk, carbon source, nitrogen source and C/N ratio as multifactor tests with the submerge culture of the mycelia to explore the effects of these various factors on the mycelial yield. The results of this experiment indicated that the optimal rotational speed for the growth of mycelia is 150 r/min, temperature 25~30 ℃, pH value 5.5~6.5, bottled volume 125 ml, and inoculated bulk 5 ml/100 ml. Meanwhile glucose is the optimum carbon source, rice bran the nitrogen source, and the C/N ratio 25:1. The optimal nutritional conditions when the mycelia of this sort of fungi were cultivated by means of submerged culture are as follow: 3% glucose, 0.072% KNO3, 0.02% NaH2PO4 and pH 8.5. Through the growth curve test the logistic equation is deduced: Y=0.616/(1+17.732e-8.874x). The weight of the dry mycelia reaches 500~700 mg/100 ml.
