桑黄子实体多糖的分离纯化及单糖组成研究

摘要: 目的:对桑黄子实体活性多糖的单糖组成进行研究。方法:桑黄子实体经水提、醇沉、冷冻干燥得水溶性多糖,经DEAE-Sepharose Fast Flow离子柱和SephacrylS-100 High Resolution凝胶柱进行分离纯化得纯多糖PIFI,气相色谱和气质联用分析多糖的单糖组成。结果:桑黄子实体多糖PIFI的单糖组成为甘露糖、葡萄糖和半乳糖,其摩尔比为0.64:1:1.94。另外还发现一种特殊的单糖3(4)-O-甲基-己糖。结论:建立了用毛细管气相色谱及气质联用测定桑黄子实体多糖单糖组成的方法,确定了桑黄多糖PIFI的单糖组成及摩尔比,还发现一种特殊的单糖3(4)-O-甲基-己糖。

Abstract: Objective: To study the monosaccharide composition of the bioactive polysaccharide extracted from the fruiting body of Phellinus igniarius. Methods: The process of isolating a water soluble crude polysaccharide was achieved by hot water extraction, filtration, ethanol precipitation and freeze-drying. The crude polysaccharide was purified to obtain PIFI by chroma- tography on DEAE-Sepharose Fast Flow column and Sephacryl S-100 High Resolution column. The monosaccharide compo- sition and molecular ratio of the PIFI were analyzed by capillary gas chromatography and mass spectrum. Results: The monosaccharide compositions of PIFI are 3(4)-O-methyl-hexasaccharide, mannose, glucose, and galactose with the molar ratio of 0.93:0.64:1:1.94. Conclusion: The determination method of monosaccharide composition of the PIFI by capillary gas chromatography and mass spectrum is established, and the results of monosaccharide composition and molar rate of the PIFI are obtained. 3(or 4)-O- methyl-hexasaccharide is present in the PIFI.
