复合酶酶法提取荷叶黄酮类化合物的研究

Optimizing Conditions for Enzymatic Extraction of Flavonoids from Lotus Leaves

摘要: 研究复合酶解法提取荷叶黄酮类化合物的最佳工艺条件。参考料液比、pH值、加酶量、温度和时间5个因素对提取率的影响,在单因素试验基础上用正交试验优化提取条件。选择70%乙醇溶液为溶剂,料液比l:20(g/mL),选取酶解pH值、加酶量、酶解温度和时间4个因素,对酶解法提取荷叶黄酮类化合物进行L16(44)正交试验,得到荷叶黄酮类化合物的最佳提取条件为pH3.5、时间3.0h、温度35℃、复合酶用量2800U/g纤维素酶+2000U/g果胶酶,荷叶黄酮提取率为3.74%。与水提法的L16(44)正交试验结果(提取率1.59%)相比较,复合酶解法提取荷叶黄酮类化合物的效果显著(P<0.01)。

Abstract: This study aimed at determining the optimum hydrolysis conditions for the extraction of flavonoids from lotus leaves. Cellulase and pectinase were used in combination for one-step hydrolysis of lotus leaves and the solvent used was 70%. The effects of five extraction conditions including solid-to-solvent ratio, pH, enzyme dosage, temperature and hydrolysis time on extraction efficiency were investigated by one-factor-at-a-time method. The optimum solid-to-solvent ratio was found to be 1:20 g/mL. pH, enzyme dosage, temperature and hydrolysis time were optimized using a 16-run orthogonal array design to be 3.5, 3.0 h, 35 ℃, 2800 U/g cellulase + 2000 U/g pectinase, respectively. Under the optimized conditions, the extraction yield of flavonoids was 3.74% compared with 1.59% for aqueous extraction technique as optimized also using a 16-run orthogonal array design (P<0.01). Hence, we concluded that enzymatic hydrolysis provides a high-yield method for the extraction of flavonoids from lotus leaves.
