高效液相色谱法测定叶黄素顺、反异构体

摘要: 为了建立高效液相色谱法测定叶黄素顺、反异构体的方法, 通过比较C18和C30色谱柱对叶黄素异构化样品的分离效果,并采用高效液相色谱-二级管阵列检测器-大气压化学电离-质谱对叶黄素顺、反异构体进行定性定量分析。该方法C30色谱柱为固定相,以甲醇-水(98:2,V/V)为流动相,在40~200ng范围内峰面积和进样量呈良好的线性关系,且具有较高的准确度和良好的重现性。该方法经济可靠,可用于叶黄素顺、反异构体的检测。

Abstract: An HPLC-DAD-atmospheric pressure chemical ionization (APCI)-MS method was proposed to quantify and qualify lutein stereoisomers. A C30 column was found to be more effective in separating lutein stereoisomers than a C18 column. The mobile phase was composed of methanol and deionized water (98:2, V/V). A good relationship between peak area and injection amount was found in the range of 40 to 200 ng. The proposed method was accurate, reproducible, economical and suitable for the determination of lutein stereoisomers.
