十个白灵菇栽培菌株的遗传多样性分析

摘要:

本研究采用RAPD 技术对10 个不同的白灵菇株系进行遗传多样性分析。经过筛选得到的4 种随机引物共扩增出52 条DNA 片段,其中多态性条带44 条,多态性位点频率为84.6%,说明通过RAPD 扩增获得的白灵菇10个株系的遗传指纹图谱多样性丰富。利用Popgen32 生物学软件计算各株系间遗传距离并构建系统进化树,在分子水平上确定了10 个白灵菇株系间的亲缘关系,可为进行更科学、准确分类提供参考依据。

Abstract:

Diversity of genomic DNA of ten cultivated strains of Pleurotus nebrodensis was analyzed by RAPD (random amplified polymorphic DNA). 52 DNA fragments were amplified from 4 selected primers, among which 44 were polymorphic bands with 84.6% of polymorphic site frequency. The result indicated that the DNA fingerprint profiles of the ten strains of Pleurotus nebrotensis are greatly diverse. Moreover, the genetic distance among strains was calculated and the phylogenetic tree was constructed by Popgen32 software. Further, the genetic relationship of the ten tested strains was identified on molecular level, which can provide important scientific basis for accurate classification of Pleurotus nebrotensis.
