崂山光皮木瓜提取物抗氧化活性研究

摘要:

为了评价崂山光皮木瓜不同提取物的抗氧化能力,以氯仿:甲醇(1:2,V/V)作为溶剂,用索氏提取法提取崂山光皮木瓜,将粗提物依次用石油醚、乙酸乙酯和正丁醇进行萃取,选用4 种不同的抗氧化评价体系:邻苯三酚自氧化法、DPPH 法、邻二氮菲- 亚铁离子-H2O2 法和还原力法对各提取物进行抗氧化实验。同时与BHT 和没食子酸(GA)两种合成抗氧化剂进行对比。结果证明乙酸乙酯提取物具有较高的抗氧化活性,在测定浓度下,其清除过氧自由基能力均比BHT 低,清除DPPH 自由基能力均比BHT 高,在50μg/ml 浓度时,高于GA;其清除羟自由基能力及还原能力在所有测定浓度下均比BHT 高,但低于GA。

Abstract:

The solvent extract of Chaenomeles sinensis (Thouin) Koehne fruits from Laoshan was prepared by extracting with a mixture of chloroform and methanol (1:2, V/V) in a Soxhlet extractor and fractionated by sequential extraction with petroleum ether, ethyl acetate and n-butyl alcohol. The antioxidant activities (AA) of the extract and its fractions including petroleum ethersoluble fraction (PEF), ethyl acetate-soluble fraction (EAF), n-butyl alcohol-soluble fraction (BF) and water-soluble residue (WR) was evaluated by four different assays, namely, the pyrogallol autoxidation, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, the 1,10-phenanthroline-Fe2+-H2O2 and the potassium ferricyanide reduction methods and compared with those of two artificial preservatives such as butylated hydroxytoluene (BHT) and gallic acid (GA). Among the extract and its four fractions, EAF exhibited the highest AA at four different concentrations. This fraction presented lower superoxide free radical scavenging activity but higher DPPH radical scavenging activity than BHT at all concentrations tested and its DPPH radical scavenging activity was also higher than that of GA at 50 μg /ml. Its hydroxyl free radical scavenging activity and reducing power both were higher than those of BHT but lower than those of GA at all concentrations tested.
