胞菌TS1138菌株L-半胱氨酸酶法合成途径的代谢控制分析

摘要: 建立了假单胞菌 TS1138菌株L-半胱氨酸酶法合成途径的动力学模型。在L-半胱氨酸合成途径中有L-半胱氨酸合成酶和L- 半胱氨酸脱巯基酶两种酶参与作用,它们的动力学模型是基于米氏动力学建立的,L- 半胱氨酸对L- 半胱氨酸合成酶有非竞争性抑制作用。通过测定L- 半胱氨酸酶法合成过程中的中间产物和终产物的浓度,利用动力学模型进行了L- 半胱氨酸合成途径的代谢控制分析。弹性系数首先可以从动力学方程中求出,根据代谢控制分析理论,流量控制系数由弹性系数求出。研究发现,在 L- 半胱氨酸酶法合成过程中,L- 半胱氨酸合成酶和L- 半胱氨酸脱巯基酶的流量控制系数存在一个交叉点。

Abstract: A kinetic model describing the pathway for the L-cysteine enzymatic synthesis by Pseudomonas sp. strain TS1138 has been developed. In the synthesis pathway, both the L-cysteine synthetase (CS) and the L-cysteine desulfhydrase (CD) were involved with the kinetic model based on Michaelis-Menten type kinetics, and also on noncompetitive inhibition of the CS by L-cysteine. From the determination of intermediates and end products during the process of L-cysteine enzymatic synthesis, with the kinetic model, metabolic control analysis (MCA) of the pathway was carried out. From the kinetic model, the elasticity coefficients and the flux control coefficients were calculated through the pathway. It was found that there is a shift in the flux control from the CS to the CD during the enzymatic reaction.
