六种食(药)用菌中—同源内源基因的发现和序列分析(英文)

Cloning and Sequencing of an Endogenetic Unknown Gene in Six Species of Fungus

摘要: 在对15种常见食(药)用菌的基因组DNA进行RAPD分析时发现,其中有六种食用菌出现同样大小为692bp的一个条带,它们是金针菇Flammulina velutipes、秀珍菇 Pleurotus geesteyanus.Singer、小平菇Pleurotu comucopiae、杏鲍菇(Pleurotus eryngii)、茶树菇Agrocybe cylindraca maire、双孢蘑菇Agaricus bisporus。我们对六个692bp的条带分别电泳纯化回收,并成功的克隆测序分析了其中四个条带,证明四个条带为高度同源的序列,为其中四种菌的同源内源基因。经过GENBANK比较,没有发现类似基因序列。最后对其进行了初步的应用研究。

Abstract: While using RAPD to screen the genomic DNA of 15 species of common edulis and medicinal fungus, a same sizeunknown gene of 692bp was found in six of them, i.e. Flammulina velutipes, Pleurotus geesteyanus.Singer, Pleurotu comucopiae,Pleurotus eryngii, Agrocybe cylindraca maire and Agaricus bisporus.These 692bp DNA fragments were collected and purified,four of them were successfully cloned and sequenced respectively, and the results indicated that these fragments’ sequencesshow a high degree of homology, they were assumed as same endogenetic unknown gene in four species of fungus. And no similargenome sequences was searched in the GenBank.
