金褐链霉菌SYAU0709中AURJ3M基因的克隆和序列分析

摘要: 以金褐链霉菌SYAU0709的DNA为模板,根据GenBank中公布的PimM的序列设计引物,通过PCR扩增方法从金褐链霉菌中扩增出约579bp的目的片段。将PCR产物克隆至PMD19-T Vector中,通过PCR扩增及测序分析发现,金褐链霉菌SYAU0709来源的AURJ3M基因和纳塔尔链霉菌PimM基因的序列具有95%的相似性。通过摇瓶发酵显示AURJ3M基因能促进金褐霉素的生物合成。

Abstract: A pair of primers were designed based on PimM gene sequences published in GenBank and a DNA fragment of approximate 579bp was amplified from Streptomyces aureofuscus SYAU0709 by PCR method. The amplified PimM gene was ligated into vector PMD19 and the recombinant Plasmid was identified by PCR amplification and sequencing. Sequence analysis indicated that the sequence of the cloned AURJ3M gene from Streptomyces aureofuscus SYAU0709 is the 95% similarity as the published PimM gene from Streptomyces natalensis.The fermentation results showed AURJ3M gene could promote Aureofuscin biosynthesis.
