薄层扫描测定大豆卵磷脂中磷脂酰胆碱的含量

摘要: 采用双波长薄层扫描法测定大豆卵磷脂中磷脂酰胆碱的含量。以氯仿:乙醇:乙酸:水为展开剂,体积比为40:9:3:1.5,测定波长为380nm,参比波长为354nm,反射法双波长锯齿形扫描磷脂酰胆碱峰面积,计算自制样品大豆卵磷脂中磷脂酰胆碱的含量。测得板间精密度为4.12%,板内精密度为2.16%,稳定性相对标准偏差为3.20%,回收率的相对标准偏差为3.26%。该方法耗时短,稳定性较好,适合于磷脂类物质的含量测定。

Abstract: The content of phosphotidylcholine in soy lecithins was determined by using dual wavelength thin-layer chromatographic scanner. The determination was conducted under the conditions of chloroform-ethanol-acetic acid-water(40:9:3:1.5,V/V) as developing solvents on a TLC silica palata,scanning wavelength 380 nm,referance wavelength 354 nnm,and dual wavelength reflection zigzay sanning the peak area of phosphatidylcholine. The results showed that the relative standard deviation(RSD) of stability is 3.20%,and the RSD of recovery rate is 3.26%. The method is simple and economical with good repetition and stability. So it could be applied in for the determination lecithin compounds.
