阳离子交换法分离纯化酵母细胞提取液中的谷胱甘肽

Glutathione Isolation and Purification of Yeast Extract with Ion Exchange Resin Columns

摘要: 谷胱甘肽是重要的生物活性物质,本文采用国产D001阳离子交换双柱(A柱和B柱)对酵母细胞提取液中的GSH进行了分离纯化,确定了A、B离子交换柱分离纯化酵母提取液中的谷胱甘肽的工艺参数为:A柱上样的提取液pH为5.5~6.0;B柱上样液(A柱的收集液)的pH为2.4~2.8;B柱的洗脱采用1.2%的NaCl溶液。A、B两柱上样和洗脱液的流速均为0.5ml/min。酵母提取液中GSH的回收率大于70%,纯化倍数大于100倍。

Abstract: Glutathione,one of the most important biological active substance plays a central role in metabolic pathways of live organisms.In this paper,A,B two cation ion-exchange columns were used to isolate and purify of GSH from yeast extract.The technology parameters of both A and B were:for column A,sample pH 5.5~6.0,for column B,sample pH 2.6~2.8,column B eluate 1.2% NaCl.The recovery rate of GSH is more than 70%,while the GSH has been purified more than 100 times.
