响应面法优化酶辅助闪式提取二氢槲皮素的工艺研究

Optimization of Homogenate Extraction for Dihydroquercetin with Cellulase by Response Surface Methodology

摘要: 目的:利用响应面法以期对纤维素酶辅助闪式提取二氢槲皮素的工艺条件进行优化。方法:以二氢槲皮素的提取率为指标,在单因素试验的基础上,应用Box-Benhnken 中心组合设计试验,对影响提取率的条件:乙醇体积分数、加酶量和液料比进行优化。结果:最佳提取条件为:乙醇体积分数为60.93%、加酶量为3.48mg/g、液料比为103.64:1 mL /g、提取时间120s。结论:闪式提取操作简便,节能,省时,可应用于二氢槲皮素大规模生产。

Abstract: Objective: Response surface analysis (RSA) methodology was used for optimizing homogenate extraction processing of dihydroquercetin with cellulose. Methods: Dihydroquercetin extraction rate as an indicator, based on single-factor experiments, the effects of ethanol concentration, amont of cellulase and liquid-material ratio on extraction rate of dihydroquercetin were explored by Box-Benhnken central composite design method. Results: The optimum extraction conditions were ethanol concentration of 60.93%, amont of cellulase of 3.48mg/g, liquid-material ratio of 103.64:1 mL /g and extraction time of 120s. Conclusion: Homogenate extraction is easy to operate, energy-saving, time-saving, it’s a method for dihydroquercetin to apply dihydroquercetin mass production.
