小麦肽对急性酒精中毒小鼠抗氧化功能的影响

Effects of Wheat Peptides on Antioxidant Function in Acute Alcoholism Mice

摘要: 摘 要:通过研究小麦肽对乙醇氧化损伤小鼠血清和肝脏组织中氧化损伤及抗氧化指标的影响,初步评价小麦肽对急性酒精中毒小鼠是否具有抗氧化功能。本研究将60只清洁级健康雄性ICR小鼠按体重随机分为5组,分别为空白对照组、模型对照组及小麦肽低、中、高三个受试物干预剂量组(0.4 g/kg bw、0.8 g/kg bw、1.6 g/kg bw),连续灌胃30天,末次灌胃后,模型对照组和三个剂量组禁食16小时,再按照12ml/kg bw 灌胃给予50%无水乙醇,空白组不作处理;6小时后摘眼球取血处死小鼠,测定血清、肝脏中总超氧化物歧化酶(total superoxide dismutase,T-SOD)活性、谷胱甘肽(reduced glutathione,GSH)含量、丙二醛(malondialdehyde,MDA)含量及肝脏组织中蛋白质羰基(protein carbonyl,PC)含量。结果表明:与模型对照组相比,小麦肽各剂量组能显著提高小鼠肝脏中T-SOD的活力(P<0.01);小麦肽低剂量组能显著提高小鼠血清和肝脏中GSH含量(P<0.05,P<0.01)、显著降低肝脏中PC的含量(P<0.05);小麦肽高剂量组能显著降低小鼠血清中MDA含量(P<0.05)。提示小麦肽能提高急性酒精中毒小鼠体内的抗氧化能力,对小鼠急性酒精性肝损伤具有一定的保护作用。

Abstract: Abstract: The effects of wheat peptides on oxidative damage and antioxidant indexes in serum and liver tissues of ethanol-injured mice were researched to preliminary study the antioxidant function of wheat peptides on acute alcoholism mice. Healthy male ICR mice were randomly divided into five groups by weight: blank control group, model control group, and wheat peptides with low, medium and high intervention dose group (0.4 g/kg bw, 0.8 g/kg bw, 1.6 g/kg bw), and gavaged for 30 days. After the last lavage, model control group and three dose groups were gavaged 50% anhydrous ethanol lasting 16 hours (12 ml/kg bw), and blank group was not processed; The total superoxide dismutase (T-SOD) activity, reduced glutathione (GSH), malondialdehyde (MDA) and protein carbonyl (PC) in serum and liver of mice were determined after 6 hours. The results showed that: compared with the model control group, the activity of T-SOD in mice liver was significantly increased in each dose of wheat peptide group (P < 0.01). In the wheat peptide low dose group, the content of GSH in serum and liver of mice was significantly increased (P < 0.05, P < 0.01), and the content of PC in liver was significantly decreased (P < 0.05). The wheat peptide high dose group could significantly reduce the content of MDA in mice serum (P < 0.05). These results suggest that wheat peptides can enhance the antioxidant capacity in mice with acute alcoholism and have a protective effect on acute alcoholic liver injury in mice.
