PI3K/AKT信号通路对冷却滩羊肉贮藏期间细胞凋亡的影响

摘要: 验证PI3K/AKT信号通路抑制的有效性,分析PI3K/AKT信号通路对冷却滩羊肉贮藏期间细胞凋亡途径的影响。以4℃条件下不同贮藏期滩羊后腿肉为研究对象,采用10 μM PI3K抑制剂LY294002溶液对滩羊肉进行注射处理,分别贮藏0 d、2 d、4 d、6 d、8 d,通过聚丙烯酰胺凝胶电泳(SDS-PAGE)和蛋白质免疫印迹(Western Blot)分析PI3K、AKT蛋白的表达情况,以此验证PI3K/AKT信号通路抑制的有效性,同时分析滩羊肉贮藏期间能量因子、氧化应激水平、线粒体损伤程度以及Caspase-3活性的变化,以探索PI3K/AKT信号通路对冷却滩羊肉贮藏期间细胞凋亡途径的诱导机制。结果表明:随着贮藏天数的延长,PI3K和AKT蛋白表达量均呈下降趋势,说明抑制剂LY294002使PI3K/AKT信号通路失活;与对照组相比,LY294002处理组线粒体ATP、糖原含量、琥珀酸脱氢酶(SDH)和柠檬酸合成酶(CS)活性显著下降(P < 0.05);线粒体ROS逐渐增多,氧化应激水平和MPTP开放程度增加;膜电位显著下降(P < 0.05);凋亡因子Caspase-3活性极显著高于对照组(P < 0.01),表明PI3K/AKT信号通路通过促进ROS产生并与PI3K相互作用,介导了下游Caspase-3的活化,使线粒体结构与功能受损,导致MPTP开放、膜电位降低,进而诱发细胞凋亡。

Abstract: Verify the effectiveness of PI3K/AKT signaling pathway inhibition, and analyze the impact of PI3K/AKT signaling pathway on cell apoptosis pathway during storage of chilled Tan mutton. The study was conducted on the leg meat of Tan mutton stored at 4℃ for different periods, using 10 μM PI3K inhibitor LY294002 solution was injected into Tan mutton and stored for 0, 2, 4, 6 and 8 days respectively. The expression of PI3K and AKT proteins was analyzed by polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot, so as to verify the effectiveness of PI3K/AKT signal pathway inhibition, and analyze the energy factors, oxidative stress levels The degree of mitochondrial damage and changes in Caspase-3 activity were investigated to explore the induction mechanism of the PI3K/AKT signaling pathway on the cell apoptosis pathway during storage of chilled Tan mutton. The results showed that with the extension of storage days, the expression levels of PI3K and AKT proteins showed a decreasing trend, indicating that inhibitor LY294002 inactivated the PI3K/AKT signaling pathway; Compared with the control group, LY294002 treatment significantly reduced mitochondrial ATP, glycogen content, SDH, and CS activity (P < 0.05); The mitochondrial ROS gradually increases, and the levels of oxidative stress and MPTP opening increase; Membrane potential decreased significantly (P < 0.05); The activity of apoptosis factor Caspase-3 was significantly higher than that of the control group (P < 0.01), indicating that PI3K/AKT signaling pathway mediated the activation of downstream Caspase-3 by promoting the production of ROS and interacting with PI3K, damaged the structure and function of mitochondria, led to the opening of MPTP, decreased membrane potential, and then induced apoptosis..
