Indirect Competitive Enzyme-Linked Immunosorbent Assay Based on Platinum Nanoparticles as Mimic Enzyme for Determination of Imidacloprid Residues in Fruits and Vegetables

摘要: 吡虫啉(imidacloprid,IMI)作为一种新型烟碱类农药,长期或过量摄入会对人体健康造成潜在危害,因此开发快速、高灵敏度的IMI残留检测技术具有重要的现实意义。基于铂纳米颗粒(platinum nanoparticles,PtNPs)的类过氧化物酶活性构建一种间接竞争酶联免疫吸附法(indirect competitive enzyme-linked immunosorbent assay,ic-ELISA),用于果蔬中IMI残留的定量检测。结果表明,PtNPs-ic-ELISA在0.1~100 μg/L范围内线性关系良好(R2=0.995),比色模式下的检出限低至0.115 μg/L。在山药、香蕉和菠菜中的平均加标回收率为91.60%~106.60%,显示出良好的准确度与适用性。该方法操作简便,具有较高的灵敏度与良好的特异性,综合性能满足我国现行标准对农产品中IMI最大残留限量的检测要求。

Abstract: Imidacloprid (IMI), as a new type of neonicotinoid pesticide, poses potential health risks to humans when ingested long-term or in excessive amounts. Therefore, developing a rapid and highly sensitive detection technology for IMI residues is of great significance for ensuring food safety. In this study, we developed a indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) based on the peroxidase-like activity of platinum nanoparticles (PtNPs) for the quantitative detection of IMI residues in fruits and vegetables. The experimental results showed that the method exhibited good linearity in the concentration range of 0.1–100 μg/L (R2 = 0.995), with a detection limit of 0.115 μg/L. The recoveries for spiked yam, banana, and spinach ranged from 91.60% to 106.60%, demonstrating good accuracy and applicability. This method is simple to operate, and demonstrates high sensitivity and good specificity. Its overall performance meets the detection requirements of the current Chinese standards for the maximum residue limit of IMI in agricultural products.
