外周-中枢-能量代谢介导玳玳花精油改善小鼠抑郁样行为

Essential Oil from Citrus aurantium L. var. amara Engl. Flowers Alleviates Depressive-like Behaviors in Mice via the Peripheral-Central-Energy Metabolism Pathway

摘要: 目的:从外周血清和中枢海马体角度探讨玳玳花精油(Citrus aurantium L. var. amara Engl. flower essential oil,CEO)对小鼠抑郁样行为的改善作用及潜在的抗抑郁机制。方法:采用气相色谱-质谱技术分析CEO主要挥发性成分。通过慢性皮质酮注射联合慢性束缚应激构建抑郁样小鼠模型,基于行为学测试和组织病理学分析评估CEO抗抑郁效果,运用转录组学与非靶向代谢组学分别分析小鼠海马体基因表达与外周血清代谢物的变化,并将CEO主要成分与筛选出的关键途径蛋白进行分子对接。结果:CEO包含40 种化学成分,能有效改善抑郁样小鼠行为学表现,并减轻其海马体和结肠组织损伤。分析海马体差异表达基因发现,CEO可能通过腺苷酸活化蛋白激酶/沉默信息调节因子1信号上调氧化磷酸化途径中的多个关键复合物的编码基因水平。此外,CEO上调了血清中吲哚-3-乙酸水平,下调了泛酸和皮质酮等多种代谢物丰度,涉及色氨酸代谢、泛酸与辅酶A生物合成、类固醇激素生物合成等途径,提示CEO可能通过调节外周能量代谢相关信号,激活海马体内的氧化磷酸化途径,从而缓解抑郁症状。分子对接表明CEO中的β-罗勒烯、橙花叔醇、乙酸香叶酯成分与氧化磷酸化途径的关键复合物能够稳定结合。结论:CEO具有明确的抗抑郁作用,可能通过激活氧化磷酸化途径进而经由“外周-中枢-能量代谢”发挥抗抑郁作用。本研究为玳玳花相关功能食品的开发提供了初步依据。

Abstract: Purpose: To investigate the ameliorative effect of Citrus aurantium L. var. amara Engl. flower essential oil (CEO) on depressive-like behaviors in mice and its potential antidepressant mechanism from the aspects of peripheral serum and central hippocampus. Methods: The major volatile components of CEO were analyzed using gas chromatography-mass spectrometry (GC-MS). A mouse model of depressive-like behavior was established via chronic corticosterone injection combined with chronic restraint stress. The antidepressant effect of CEO was evaluated by behavioral tests and histopathological analysis. Transcriptomics and untargeted metabolomics were used to analyze changes in hippocampal gene expression and peripheral serum metabolites in mice. Molecular docking was performed between the major components of CEO and the key pathway proteins. Results: CEO, containing 40 chemical components, effectively improved the behavioral performance of depressive-like mice and alleviated damage to their hippocampal and colonic tissues. Analysis of differentially expressed genes in the hippocampus revealed that CEO may upregulate the expression of genes encoding multiple key complexes in the oxidative phosphorylation pathway via the adenosine monophosphate-activated protein kinase (AMPK)/silent information regulator 1 (SIRT1) signaling pathway. In addition, CEO increased the serum level of indole-3-acetic acid and decreased the abundance of various metabolites such as pantothenic acid and corticosterone, which are involved in pathways including tryptophan metabolism, pantothenic acid and coenzyme A biosynthesis, and steroid hormone biosynthesis. These findings suggest that CEO may alleviate depressive symptoms by regulating peripheral energy metabolism-related signals to activate the oxidative phosphorylation pathway in the hippocampus. Molecular docking indicated that β-ocimene, nerolidol, and geranyl acetate present in CEO could bind stably to the key complexes in the oxidative phosphorylation pathway. Conclusion: CEO exerts a definite antidepressant effect, which may be mediated through activating the oxidative phosphorylation pathway and further via the “peripheral-central-energy metabolism” pathway. This study establishes a preliminary basis for the development of functional foods based on C. aurantium L. var. amara Engl. flower.
