Study on Extraction of Androst-4-diene-17-dione from Oil-water Two-phase Fermentation System of Mycobacterium B696

Abstract:

Mycobacterium B696 can effectively degrade soybean sterol to androst-4-diene-17-dione (4-AD) in oil-water twophase system, but how to enhance the extraction rate and purity of 4-AD is still a problem at present. In the present study, the effects of different pretreatments, extraction ways and crystallization on the extraction rate were investigated. The results showed that the optimal extraction technology was as follows: after 10 min of heat treatment at 50 ℃, extracting the oil phase with 3 times volume of methanol at 15℃, and then conducting the crystallization of 4-AD at 10 ℃ for 15 h. Under these conditions, the extraction yield of 4-AD was 76.61%, and its purity was up to 98.24%.
