Abstract: Objective: To establish optimal parameters for the separation and purification of total flavonoids from the whole plant of Stenoloma chusana (L.) Ching using AB-8 macroporous adsorption resin. Methods: Static and dynamic adsorption and desorption behaviors of AB-8 macroporous adsorption resin were evaluated for achieving the best purification of total flavonoids. Results: The optimum adsorption and desorption parameters were found to be: sample pH 4.5, sample concentration 1.00 mg/mL, sample loading flow rate 80 mL/h, 70% ethanol as the eluent, desorption flow rate 40 mL/h and eluent amount 60 mL, resulting in a total flavonoid purity of 66.16%. Conclusion: AB-8 macroporous adsorption resin is safe, economical and easy to use and can therefore be promisingly applied to separate and purify total flavonoids from the whole plant of Stenoloma chusana (L.) Ching.
