Selection and Enzymetic Study of an Agarolytic Bacterium AT-22

Abstract: The agar-decomposing strain AT-22 was isolated and selected from some species of red algae( Gelidium amansii) in the coastal water of Qingdao,and presumptively classified within the genus Pseudomonas. Its growing and enzymeology properties were studied primarily. The results show that the agarase produced by AT-22 is an inducible enzyme, and the enzyme producing ability will be restrained by addition of 0.2% Glucose in culture medium. The optimal pH is at 6.0~7.0 and temperature 40℃, the optimal concentration of substrate is 1.0%~1.2%,addition of Ca2+ will greatly promote the enzyme activity,whereas Fe3+, Mn2+, Cu2+ and Hg2+ cause inhibition.
