Preliminary Investigation into the Separation of Lutein and Related Compounds from Marigold Flower by C30-HPLC-PDA

Abstract:  The lutein and related compounds from the flower of marigold were readily separated by C30-HPLC-PDA,including the separation of lutein esters and geometrical isomers,under following conditionWith YMCTM Carotenoid S-5 column (250 ×4.6mm),mobile phase A and B were acetonitrile-methanol (75:25,V/V) and MTBE ,respectively. Linear gradient elution was performed at the flow rate of 1.0 ml/min. The PDA acquired the spectra of fractions from 300 to 600nm. Sample injection volume was 20μl. According to its chromatographic behavior,spectral characters and geometrical transition catalyzed by iodine,each fraction was preliminarily identified. Result from this study suggested that the application of C30-HPLC-PDA in the separation of lutein and related compounds from marigold flower exhibited a bright future.
