Study on Flavonoid Assay in Lithhocarpus polgstachrch Rehd with Solid Phase Extraction and HPLC

Abstract: A high performance liquid chromatography (HPLC) method for the flavonoid assay in Lithhocarpus polgstachrch Rehd was studied. The flavonoid was extracted from the sample with 80% of ethanol by heated reflux. Then the extract was degreased by solid phase extraction with Sep-Pak-C18 cartridge. The flavonoid was separated on a Nova-Pak-C18 chromatogram column (3.9×150mm, 5μm), with methanol and 0.05mol/L potassium dihydrogen phosphate buffer solution as mobile phase at 60:40 (V/V), and monitored with the photodiode array detector at 360nm. The recoveries of the flavonoid are 97%~103%, and relative standard deviations are 0.87%~2.2%.
