Application of RAPD in Genetic Relationship Analysis of Pleurtus ostreatus Strains

Abstract: Five cultivated strains of Pleurotus ostreatus from different areas of China were assessed for their DNA polymorphism by RAPD technique.A total of 107 RAPD bands were amplified by eight selected primers,in which 69 bands were polymorphic and the rate was 64.4%.The variations of RAPD band patterns and DNA similarity among the strains revealed that there is definite genetic diversity in the natural germplasm of Pleurotus ostreatus in China.A cluster dendrogram of these samples showed that the five strains were classified into 2 groups using similarity of 0.470 as a cut-off point.Group1 consisted of PL-72,PL-2,Yanping 2106 and PL-A2,and group two was PL-AD28.While using 0.570 as a cut-off point,the group 1 was further classified two subgroups.One was comprised of PL-72 and Yanping 2106,and the other included PL-2 and PL-A2.This study provide molecular evidence for selection and breeding of eminent cultivars and genetic relationship in Pleurotus ostreatus.
