Study on Mutation Breeding of High Methionine Producing Strain from Corynebacterium pekinense AS1.299

Abstract: Corynebacterium pekinense AS1.299 was mutagenized with ultraviolet(UV) and N-methyl-N’-nitro-nitrosoguanidine(NTG) to screen high methionine-producing strain in this study.Results showed that for the Corynebacterium pekinense AS1.299 suspension,the optimal conditions of UV mutagenesis are bacterial concentration 103 CFU/ml and UV exposure time 18 s,and the optimal conditions of NTG mutagenesis are NTG concentration 102 CFU/ml and treatment time 40 min.After three-circle alternation of UV and NTG mutagenesis,the methionine productivity of the mutant strain is 1.103 g/L,0.703 g/ L higher than that of original strain.After five passagings,its genetic stability is still good.
