Coexpression of Glycerol Dehydrogenase Gene (dhaD) and Dihydroxyacetone Kinse Gene (dhaKL)

Abstract: The gene of dhaD encoding glycerol dehydrogenase was amplified by PCR from the genomic DNA of Klebsiella. pneumoniae As1.1736 and the expression vector pET-28a(+)/dhaKL-dhaD was constructed. Sequence analysis showed that dhaD gene is 1104 bp length and encodes 367 amino acid residues; SDS-PAGE showed that the dhaKL gene and dhaD are expressed, and the enzyme activities of glycerol dehydrogenase and dihydroxyacetone kinase are 23.2 U/ml 25.6 U/ml respectively in supernant.
