Synthesis and Immunological Potency Determination of Benzoic Acid Artificial Antigen

Abstract:

Through the ammonolysis method of phthalic anhydride (PA), the derivative of benzoic acid (BA) was used to synthesize the artificial antigen of BA in the present study. BA was linked to ovalbumin (OVA) to generate immunogenic antigen BA-OVA and also was linked to bovine serum albumin to generate coating antigen BA-BSA. Artificial antigen was detected and identified by UV and IR spectroscopies as well as SDS-polyacrylamide gels electrophoresis. BA-OVA was further used to immune BALB/c mice, and the generation of antibody was detected. The detection results showed that the artificial antigen of BA has been synthesized successfully. It was calculated that the ratio of BA to OVA is 174:1 based on their UV absorbance. The antibody is positive in serum of immuned mice detected by ELISA (The titration of antiserum is 1:3.2 × 105), which suggests that the antigen can induce the generation specific antibody of BA. The synthesis of BA artificial antigen makes it possible to establish monoclonal antibodies.
