Optimization of Extraction Process for Flavonoids from Polygonum aviculare L. Based on HPLC Quantitation

Abstract: Objective: To optimize the extraction condition of flavonoids from Polygonum aviculare L based on HPLC quantitation. Methods: The effects of raw material particle size, ethanol concentration, soaking (extraction) time, solid-to-solvent ratio and extraction temperature on the extraction rate of total flavonoids were explored by orthogonal array design. Results: The optimal extraction conditions were particle size of 40 mesh, solid-to-solvent ratio of 1:30 (g/mL), extraction temperature of 80 ℃, extraction time of 2 h and ethanol concentration of 65%. The total flavonoids concentration of the extract obtained under these conditions was 228.9μg/mL. Among all tested process parameters, raw material particle size had the biggest effect on the extraction rate of total flavonoids. Conclusion: HPLC provides a reliable method for quantifying total flavonoids. The optimize extraction process provides a reliable, simple and useful approach for the extraction and exploitation of flavonoids from Polygonum aviculare L.
