Determination of Cholesterol in Milk by HPLC

Abstract: Objective: To establish an HPLC method for the determination of cholesterol in milk. Methods: Cholesterol was separated on a Shimadzu C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile as mobile phase at a flow rate of 1.0 mL/min. The detection wavelength was set at 203 nm and the column temperature at 30 ℃. Results: A calibration curve was established revealing a good linear relationship in the range of 3.58-358μg/mL with a correlation coefficient of 0.9999. The average spike recovery rate was 101.8% with a RSD of 1.14%. Conclusion: The method was accurate, reproducible and applicable for the determination of cholesterol in milk.
