Mutation Breeding of Strain Highly Producing Androst-4-ene-3,17-dione from Rapeseed Sterols

Abstract:

The mutagenesis of Mycobacterium sp. BD-696 was induced by UV and DES, and two mutants Mycobacterium sp. BD-696-3 and Mycobacterium sp. BD-696-6 were obtained. When the concentration of substrate steroid is 6‰ and shaking flask fermentation is kept for 168 hours, the transformation yield of androst-4-ene-3,17-dione (AD) from rapeseed sterol by Mycobacterium sp.D-696-3 and Mycobacterium sp. BD-696-6 are 2.31 g/L and 2.33 g/L, respectively, and the corresponding transformation rates are 60.7 % and 61.4%, higher 12.8% and 13.5% than that by the original strain, respectively. Moreover, the continuous passage experiment showed that the mutant BD-696-6 has high genetic stability.
