Methodology Study of Indirect Competitive ELISA for Detection of Vomitoxin

Abstract: Objective: An indirect competitive ELISA was developed by using anti- DON monoclonal antibody and coating antigen DON-BSA for detection of deoxynivalenol in wheat and corn. Methods: The best dilute concentrations of anti-DON monoclonal antibody and coating antigen were obtained by matrix titration experiment. Methodology of ELISA kit was also studied to ascertain qualifications. Results: The detection limits was 10μg/L. and the linear range of the inhibition curve was 10~500μg/L. The mean coefficients of variation was 5.8%, and mean recovery rate of DON in the samples was 104%. The detection could be finished within 2h. The kit has more than six months stationery phase during 4℃ storage. Conclusion: This ELISA method was verified to be rapid, safe, sensitive, repeatable, specific for DON detection.
