Study on Optimization of the Working Conditions for Extracting Carotenoids from Azotobacter-strain β-C1

Abstract: The cell-disrupting methods, solvent extracting and working conditions for extracting carotenoids form theAzotobacter strainβ-C1 were studied. The method of hot hydrochloric acid was the best to bread the cell wall, and the optimalconditions obtained by orthogonal experiment were: concentration of HCl 3mol/L, HCl addition 30ml/g(dry cell), temperature100℃ and time 2min .Ethanol was the suitable solvent as far as the toxin problem and application convenience were concerned.Besides the optimal conditions were: alcohol addition 30ml/g (dry cell), extracting temperature 50℃ and extracting time 45min.Under the optimal conditions for disrupting cell and extracting carotenoids, the extraction rate was 3.45mg/g.
