Separation and Purification of Adenosine 3′,5′-Cyclic Monophosphate(cAMP) from Hetian Jade Jujube

Abstract: Xinjiang hetian jade Chinese jujube was used as the raw material to separate cAMP,and then it was purified with several resins. It was found that LS-200 resin is the most suitable one. The optimal technical parameters of absorbing cAMP dynamically are as follows:cAMP sample volume 8 times of LS-200 resin,its pH 4,flow rate 1 ml /min,and concentraction 30μg/ml,and those of desorbing cAMP dynamically are as follows:taking 35% ethanol as eluent,its flow rate 1 ml /min,and its amount 4.8 times of LS-200 resin. The cAMP purity of freeze-dried sample after resin purification is 3.47% and it is stable at pH 4.
