Optimization of Induction Conditions of Loose Callus from Eucommia Ulmoidues

Abstract:  Callus from Eucommia ulmoides grows too rigidly to suspension culture,so rapidly vegetal and looses callus is the basis of it’s cell suspension culture. The sterile seedling as explants,the effects of different explants,concentration of NAA,2,4-D and 6-BA and 3 basic medium on Eucommia ulmoides callus induction and growth were investigated. The results showed that the epicotyl and cotyledon are much easier and the MS medium is the best medium for callus induction. The results of L9(34) orthogonal test indicated that properly increasing 6-BA content could reduce darkening and improve loose callus formation,and the optimum hormone combination was 1.0mg/L2,4-D+1.0mg/LNAA+1.0mg/6-BA.
