Identification of A Microorganism Strain Producing the Antimicrobial Substance with 16S rDNA Sequence Analysis

Abstract: A microorganism strain producing the antimicrobial substance was isolated from peaches and was named fmb3. The inhibitory test showed that the antimicrobial substance produced by fmb3 can strongly inhibit the gram positive strains Staphy lococcus aureus and Bacillus cereus,the gram negative strain Escherichia coli and the mold Penicillium expansum. Identification of the microorganism strain was also performed using the 16S rDNA sequence analysis. First the genomic DNA from fmb3 was extracted,and then the general primers were designed according to conservative sequence of bacteria 16S rDNA. Furthermore,amplifying the target 16S rDNA sequence and the sequence analysis by using the NCBI BLAST database was described. The strain was finally identified as the Bacillus subtilis.
