Exploration of loop-mediated isothermal amplification (LAMP)for detection of alicyclobacillus acidoterrestris

Abstract: [Purposes] A Loop-mediated isothermal amplification (LAMP) method was established for detection of Alicyclobacillus acidoterrestris in food. [Methods] After optimization of the reaction conditions of LAMP, including the concentrations of primers, reaction time, and the amplification temperature, Alicyclobacillus acidoterrestris -LAMP method was assembled, meanwhile sensitivity and specificity were evaluated in this paper. [Results] The results showed that the method was available to detect Alicyclobacillus acidoterrestris in foods. The Alicyclobacillus acidoterrestris could be rapidly and specifically detected by LAMP method within 1 hour at constant temperature of 63℃. The sensitivity of the method to detect Alicyclobacillus acidoterrestris was at the level of 6.7 CFU/mL and the specificity of the approach was 100%. [Conclusions] The Alicyclobacillus acidoterrestris -LAMP method is effective, high sensitivity and specificity for the detection of Alicyclobacillus acidoterrestris, and available for screening of Alicyclobacillus acidoterrestris in a large number of food samples.
