Toxicological Safety and Prebiotic Function Evaluation of Konjac Oligosaccharide

Abstract: To evaluate the toxicological safety and Prebiotic function of konjac oligosaccharide(KOS). Methods:mouse acute toxicity test, mouse bone marrow cell micronucleus test and mouse sperm aberration test were used to evaluate its toxicological safety, the Prebiotic function of was evaluated by in vitro fermentation of mouse cecum contents. Results: Acute toxicity test showed that the median lethal dose of KOS to male and female mouse were both more than 21.5 g/kg body weight, indicating KOS was non-toxic. The results of genotoxicity tests, including mouse bone marrow cell micronucleus test, mouse sperm aberration test, were all negative. in vitro fermentation showed that KOS could promote the proliferation of beneficial bacteria such as Bifidobacterium spp. and Lactobacillus/Enterococcus spp,but made no significant difference on Escherichia Coli. In addition,KOS could significantly promote the accumulation of acetic acid, propionic acid ,butyric acid and valeric acid generated by gut microbes, and then improve the total amount of short-chain fatty acid (SCFAs). Conclusion:The KOS has high safety and an excellent prebiotic function.
