Optimization of Ultrasonic-Assisted Extraction Conditions of Yohimbine from Rauvolfia by Response Surface Analysis

Abstract:   The ultrasound extraction process parameters of yohimbine from Rauvolfia in Guangxi province was optimized by response surface methodology. Four independent influencing factors of extraction, such as comminution size of the medicine material,pH, extraction time and the ratio of liquid to solid were considered on the basis of single factor experiments.And then the experiments were arranged according to Box-Behnken central composite experiment design and response surface analysis. The results showed that the optimum ultrasound extraction conditions for yohimbine from Rauvolfia were found to be 120 mesh comminution size, 18:1 solvent/solid (mL/g) , pH 1, 59.75 min. Under the optimized conditions, the yohimbine yield was 3.841mg/g. And It was in good agreement with its predicted value (3.9038mg/g) by model.The optimum ultrasound extraction conditions was reliable, and it could be used to guide the actual production.
