Separation, Purification and Monosaccharide Composition of Polysaccharides from the Ink of Sthenoteuthis oualaniensis in South China Sea

Abstract: Objective: To isolate and purify polysaccharides from the ink of Sthenoteuthis oualaniensis in the South China Sea, and to analyze the monosaccharide composition of the purified polysaccharides. Methods: Crude polysaccharides were isolated through water extraction followed by alcohol precipitation and passed through a solid phase extraction column to remove the pigment. The polysaccharides were fractionated and further purified into homogeneity by sequential chromatographies on DEAE-52 ion exchange and Sephacryl HR-300 columns. The monosaccharide composition was analyzed by ultra performance liquid chromatography (UPLC) after 1-phenyl-3-methyl-5-pyrazolone (PMP) derivatization. Results: Three purified polysaccharides designated as SIP1, SIP2, and SIP3 were obtained. SIP1 was composed of D-mannose, D-glucose, and D-galactose while SIP2 and SIP3 were composed of D-mannose, rat sugar, D-glucose, D-galactose and L-(-)-fucose in different ratios.
